Al Madrigal was born on 4 July 1971 in San Francisco, California. Madrigal is of Sicilian and Mexican descent. He grew up in San Francisco, where Mike Pritchard and Margret Cho were his neighbors. Madrigal began his studies at Ecole Notre Dame Des Victories, which is a catholic private school where the French culture and language are mandatory.

Madrigal began his career in comedy as a stand-up comedian in 2002. He performed as a solo performer and also in a group that he had co-founded by the name Fresh Robots. In 2002 Madrigal performed at both SF Sketchfest comedy festival and New Faces, which was a showcase of Just for Laughs festival. Madrigal signed a talent holding agreement in 2004 with CBS after he had won a Jury award for the best stand-up comedian at the US comedy arts Festival. His half-hour comedy special titled Madrigal’s Comedy Central aired in 2005.
In 2003, he was to play in a comedy series The Ortegas, but the show got canceled before it got released. He played the role of a building attendant by the name Jesus in the comedy series Welcome to the Captain in 2008. However, the series got canceled after five episodes. He then starred in the series Gary Unlimited which began airing on TV in 2008.
In 2011, it was publicized that Madrigal would join the cast of The Daily Show with Jon Stewart. During his audition, for the show, he performed a piece that he had played for Debbie Riddle, a tax representative who had proposed a bill whereby employers would get punished for hiring immigrants.
